猪链球菌2型毒力相关蛋白的检测及其基因片段的克隆与表达

136kD的溶菌酶释放蛋白(muraminidase-releasedprotein,MRP)和110kD的胞外蛋白因子(extracellular proteinfactor,EF)是猪链球菌2型(Streptococcus suis type 2)的毒力相关蛋白。提取猪链球菌2型江苏分离株HA9801的胞壁蛋白和胞外蛋白,分别进行SDS-PAGE,切割相对分子量为136kD的MRP和110kD的EF,按常规方法制备其抗体。用此抗体分别以免疫转印和ELISA法检测猪源链球菌18株国内分离株、1株德国分离株及1株猪链球菌2型人分离株的毒力相关蛋白。提取20株待检菌株的胞壁和胞外蛋白,经测SDS-PAGE,与HA9801株的MRP和EF的抗体作免疫转印。结果显示,11株MRP阳性,10株EF及EF~*阳性,MRP及EF的分布存在4种表型:MRP~+EF~+(8/20),MRP~+EF~*(1/20),MRP~+EF~-(1/20)  (本文共114页) 本文目录 | 阅读全文>>

两种猪源链球菌纤连蛋白体外结合的特征

猪链球菌是集约化养猪场的重要的病原之一。迄今为止,发现有35个血清型。猪链球菌2型(Streptococcus suis 2,SS2)被认为是毒力最强的。马链球菌兽疫亚种(Streptococcus equi subsp.zooepidemicus,SEZ)可感染猪、马、犬、羊、鸡、猫及奶牛,在兽医方面具有重要的意义。上述两种菌均是人兽共患病病原,也是我国猪链球菌病的主要病原。细菌表面的疏水性是细菌重要的理化特性,与细菌表面组成物质有很大关系,一定程度上与细菌的致病性有关。纤连蛋白(fibronectin,Fn)是由α和β链组成的一种二聚体糖蛋白,相对分子质量(Mr)约450kD,由多种类型的细胞表达产生,在动物体内两种存在方式,一种游离存在于血浆中,一种以纤维状存在于细胞外基质(ECM);α与β链中均含有Ⅰ型、Ⅱ型和Ⅲ型三种重复序列;整合素、肝素、胶原/明胶和血纤蛋白都与Fn结合,位置各不相同。细菌表面的Fn结合蛋白在Fn上有...  (本文共103页) 本文目录 | 阅读全文>>

猪链球菌防治新技术研究

本研究对16起疑似猪链球菌感染病料进行分菌鉴定后,确认14起为猪链球菌感染,其中2型3起,7型5起,6起是1、2、7和9型之外的血清型,一起弓形虫感染,一起未查明病因。对甘、青、宁三省的猪链球菌2型的血清学调查结果表明,临床健康猪群的猪链球菌2型抗体阳性率为0~16.7%;连续5年从甘肃武威健康猪只采取扁桃体分菌,猪链球菌的分离率为27.2%,其中2型5.5%,9型13.2%, 7型11.4%,1型3.7%,66%是1、2、7和9型之外的血清型。药敏检测表明多重耐药性菌株占分离菌株的84%以上。通过基因筛选、不相合性检测及不同基因序列组合分型,发现由dnaK-EFTu-potA-eno-uvrA-guaB的基因合并序列所构建的系统发生树在各节点处的自举检验值均≥85%,该系统发育树从根本上提高了链球菌分型结果的可靠性。据此,链球菌分为3群,第一群为mitis群链球菌、猪链球菌和热链球菌,猪链球菌与mitis群链球菌的遗传关系极为...  (本文共87页) 本文目录 | 阅读全文>>
