螺内酯对百草枯中毒大鼠肺纤维化的干预作用及其作用机制的实验研究

【实验目的】本研究通过一次性灌胃建立百草枯(Paraquat, PQ)中毒大鼠肺纤维化模型,观察螺内酯能否阻抑PQ中毒大鼠肺纤维化,以揭示PQ中毒致肺纤维化部分机理及初步探讨其可能的干预机制。【实验方法】63只健康成年雄性SD大鼠随机分为三组:正常对照组(Control组)、百草枯染毒组(Paraquat, PQ组)、百草枯+螺内酯治疗组(Paraquat +Spironolactone, PQ+SPI组)。各组大鼠按观察时点7d、14d、28d随机又分成三个亚组,每亚组7只。PQ组和PQ+SPI组大鼠于实验开始一次性PQ(80mg/kg)灌胃染毒而Control组给予2ml生理盐水灌胃。灌胃后2小时,PQ+SPI组大鼠每日予以螺内酯(Spironolactone,100mg/kg)1次灌胃干预,而PQ组和Control组则予以生理盐水2ml灌胃。各组大鼠分别于造模后第7d、14d、28d处死,处死前开胸获取血及肺组织,记录各时...  (本文共58页) 本文目录 | 阅读全文>>
