ATRA增强HSV-TK/GCV对膀胱癌旁观者效应的实验研究

研究背景:膀胱癌是泌尿系统常见的恶性肿瘤,术后高复发率和对化疗药物的不敏感一直影响着膀胱癌的疗效。单纯疱疹病毒胸苷激酶(HSV-TK)基因转染治疗膀胱肿瘤是目前肿瘤基因治疗的重要策略之一,显示了较好的研究价值和临床应用前景。实验已证实“旁观者效应”即转染阳性细胞使未转染细胞凋亡是TK基因治疗另一重要作用途径,它与基因转染效率被看成肿瘤自杀基因治疗成功的两大关键要素。在目前基因转染效率还不尽如意的情况下,旁观者效应的作用越来越受重视,探讨如何提高旁观者效应是很有意义的。许多实验已证实缝隙连接细胞间通讯(GJIC)是实现旁观者效应主要机理之一。相邻细胞膜上的连接蛋白构成的连接子相互锚定形成中空的连接通道是细胞间通讯的结构基础。然而,在许多肿瘤中,组织特异性连接蛋白表达异常,引起细胞间通讯功能下降,这是自杀基因“旁观者效应”在许多肿瘤中不明显的原因之一。国外许多实验已证实,膀胱移行上皮主要表达连接蛋白Cx26,在调节移行细胞增殖和分化  (本文共59页) 本文目录 | 阅读全文>>
