人乙醛脱氢酶2基因的克隆及表达

人体中的乙醇脱氢酶(alcohol dehydrogenase,ADH)及乙醛脱氢酶(aldehyde dehydrogenase,ALDH)在乙醇代谢过程中发挥着重要的作用。若ADH及ALDH在人体内均充分具备,则能较快得分解乙醇,减少究竟对中枢神经的影响,因而即使摄入一定量的乙醇,其所含有的乙醇也能在很短时间内得到充分的代谢。在人体中,ADH在一般情况下是可充分表达,且表达量基本持平的。人群内缺乏ALDH的个体则较多,而ALDH的缺乏将导致乙醇不能被完全分解为水和二氧化碳,而是以乙醛的形式继续留存于体内,使人在酒后产生恶心欲吐、昏迷不适等中枢神经麻痹状态,即醉酒症状。其中的ALDH-2就是众所周知的有高度遗传多态性并与亚洲人群的饮酒行为密切相关的基因。近年来,随着人们对获得全新的、更有效的解酒保肝药物的需求日益迫切,关于ALDH的研究工作更显其深远意义。本文采用PCR技术,克隆获得了人aldh2基因,构建质粒pUCm-ald  (本文共72页) 本文目录 | 阅读全文>>

乙醇脱氢酶基因和乙醛脱氢酶基因的克隆及表达

乙醇脱氢酶(ADH)和乙醛脱氢酶(ALDH)是以烟酰胺腺嘌呤二核苷酸为辅酶,分别催化醇和醛反应的酶类。在工业生产和医学应用上具有可观的研究价值。近年来由于有机相催化和膜反应器的研究发展对ADH应用起到的推动作用,以及乙醛脱氢酶在相关疾病检测方面的广泛应用,国内外学者对ADH和ALDH的关注度逐渐提升。ADH及ALDH普遍存在于动物体内和微生物中,目前国内外已有不少学者对微生物中ADH和ALDH的分离纯化进行了研究,但从动物肝脏和微生物中提取ALDH和ADH存在产量低、成本过于昂贵等问题。本文通过分子生物学手段,克隆获得乙醇脱氢酶基因及乙醛脱氢酶基因并实现高效表达,以期大量获得ADH和ALDH,主要研究结果如下:1.通过PCR方法成功获得酵母中adh2和aldh6基因,将目的基因与T载体连接后测序,经比对后确定与已公布序列相似性分别为96%和99%,序列结果上传Genbank后获得登录号JX901290、JX901291。2.利用...  (本文共66页) 本文目录 | 阅读全文>>
